چکیده

In this study, a simple and efficient metaphase chromosome preparation method from wheat was described which can also be applied on most other plants. The act of not using hydrolyzing chemicals such as hydrochloric acid in this method made the samples suitable for C-banding and in situ hybridization as well. Squeezing out the meristematic cells from the root tip region on to the surface of the slide is the main step of the described method, leading to monolayer and clean preparations. In order to arrest cells in metaphase, colchicine, -monobromonaphthalene, ice cold water, colchicine-ice cold water and -monobromonaphthalene-ice cold water were compared to each other. Ice water pretreatment resulted in the highest metaphase index (up to 20% in some cultivars). Some preparations were used for in situ hybridization. The described method can be reliably applied in the laboratory with only basic equipments.

45S rDNA Regions Are Chromosome Fragile Sites Expressed as Gaps In Vitro on Metaphase Chromosomes of Root-Tip Meristematic Cells in Lolium spp

BACKGROUND In humans, chromosome fragile sites are regions that are especially prone to forming non-staining gaps, constrictions or breaks in one or both of the chromatids on metaphase chromosomes either spontaneously or following partial inhibition of DNA synthesis and have been well identified. So far, no plant chromosome fragile sites similar to those in human chromosomes have been reported....

Molecular cytogenetic identification of a wheat (Triticum aestivum)-American dune grass (Leymus mollis) translocation line resistant to stripe rust.

Leymus mollis, a perennial allotetraploid (2n = 4x = 28), known as American dune grass, is a wild relative of wheat that could be useful for cultivar improvement. Shannong0096, developed from interspecific hybridization between common wheat cv. Yannong15 and L. mollis, was analyzed with cytological procedures, genomic in situ hybridization, stripe-rust resistance screening and molecular m...

A fluorescence in situ hybridization system for karyotyping soybean.

The development of a universal soybean (Glycine max [L.] Merr.) cytogenetic map that associates classical genetic linkage groups, molecular linkage groups, and a sequence-based physical map with the karyotype has been impeded due to the soybean chromosomes themselves, which are small and morphologically homogeneous. To overcome this obstacle, we screened soybean repetitive DNA to develop a cock...
